Structural organization of the 16S ribosomal RNA from E. coli. Topography and secondary structure.

چکیده
Extensive studies in our laboratory using different ribonucleases resulted in valuable data on the topography of the E.coli 16S ribosomal RNA within the native 30S subunit, within partially unfolded 30S subunits, in the free state, and in association with individual ribosomal proteins. Such studies have precise details on the accessibility of certain residues and delineated highly accessible RNA regions. Furthermore, they provided evidence that the 16S rRNA is organized in its subunit into four distinct domains. A secondary structure model of the E.coli 16S rRNA has been derived from these topographical data. Additional information from comparative sequence analyses of the small ribosomal subunit RNAs from other species sequenced so far has been used.

Mutational robustness of 16S ribosomal RNA, shown by experimental horizontal gene transfer in Escherichia coli.
The bacterial ribosome consists of three rRNA molecules and 57 proteins and plays a crucial role in translating mRNA-encoded information into proteins. Because of the ribosome's structural and mechanistic complexity, it is believed that each ribosomal component coevolves to maintain its function. Unlike 5S rRNA, 16S and 23S rRNAs appear to lack mutational robustness, because they form the struc...
متن کاملStructure-function relations in E. coli 16S RNA.
As a more thorough understanding of RNA secondary and tertiary structure has developed over the last several years, so has an appreciation of its importance in the function of the various RNP particles of which it is a part. For example, Ul RNA has been linked to mRNA splicing (Lerner et al., 1980; Rogers and Wall, 1980) while the RNA moiety of RNAase P has been shown to be absolutely required ...
متن کاملAn experimentally-derived model for the secondary structure of the 16S ribosomal RNA from Escherichia coli.
Ribonucleoprotein fragments are isolated by mild ribonuclease digestion of E. coli 30S ribosomal subunits, and are deproteinized and subjected to a second partial digestion. Base-pairing between the resulting small RNA fragments is investigated using the two-dimensional gel electrophoresis procedure already reported (see Ref. 1). The interactions thus found are incorporated into a secondary str...
متن کاملDevelopment of 16S rRNA targeted PCR methods for the detection of Escherichia coli in Rainbow trout (Oncorhynchus mykiss)
Objectives: The presence of E.coli in fish intended for human consumption may constitute a potential danger, not only in causing disease, but also because of the possible transfer of antibiotic resistance from aquatic bacteria to those infecting humans. The objective of this study was to develop an improved PCR method based on species – specific 16 S rRNA gene primers (FES,...
متن کاملThe Escherichia coli ribosomal RNA leader nut region interacts specifically with mature 16S RNA.
All ribosomal RNAs are preceded by leader sequences not present in the final ribosome particles. The highly conserved leader sequences of bacterial rRNAs are known to be important for the folding and assembly of functional ribosomes. Very likely transient binding of the leader to mature parts of the 16S RNA occurs during transcription. To better understand the mechanistic details of these funct...
